Regulación génica durante el desarrollo y regeneración de las células beta pancreáticas

La patología diabética se asocia con la muerte o mal funcionamiento de las células beta productoras de insulina del páncreas. Estudios recientes han demostrado una insospechada capacidad de regeneración del páncreas adulto en roedores, siendo capaces de revertir el estadio diabético en respuesta al daño tisular. Nuestro laboratorio estudia los mecanismos moleculares involucrados en el control de la generación y regeneración de las células beta pancreáticas.

La patología diabética se asocia con la muerte o mal funcionamiento de las células beta productoras de insulina del páncreas. Estudios recientes han demostrado una insospechada capacidad de regeneración del páncreas adulto en roedores, siendo capaces de revertir el estadio diabético en respuesta al daño tisular. Nuestro laboratorio estudia los mecanismos moleculares involucrados en el control de la generación y regeneración de las células beta pancreáticas. A estos fines integramos el análisis bioinformático de datos epigenómicos (ChIP-seq) y transcriptómicos (RNA-seq) obtenidos en muestras pancreáticas humanas, de rata y de ratón, con la validación experimental en modelos biológicos apropiados. Usando este enfoque, reportamos previamente que las proteínas TEAD y YAP regulan la red de regiones regulatorias distales del genoma (o enhancers, por su término en inglés) que está activa durante el desarrollo pancreático en humanos. Actualmente estamos estudiando como los glucocorticoides podrían interferir con este mecanismo. Por otro lado, nuestros resultados mas recientes están orientados al estudio de los mecanismos involucrados en la regeneración de células beta en roedores, poniendo en un plano central a la proteína INGAP ( “Islet Neogenesis Associated Protein”) y a un potencial rol del sistema inmune en este proceso.
